"Complete item designation" is typically used as an alternative to a combination of the other options here, which are connected to one aspect (= function, + location, - product) each. The complete item designation is typically a combination of multiple aspects, like for instance =A1=B5+R3+C1-K1.
If it is a good idea or not to include parts of the complete item designation in the definition of numbering areas, depends on the overall project structure. Please refer to the General orientation about numbering areas topic for a further explanation about that.
Please note, that when we talk about "levels" for a complete item designation, we count them from left to right, regardless of aspect. Please refer to the table below, which shows an explanatory example.

The first level of the complete item designation, counted from left to right, is included in the numbering area definition. Any lower levels, if they exist, are not included.

2 - The two first levels only |
The two first levels of the complete item designation, counted from left to right, are included in the numbering area definition. Any levels lower than that, if they exist, are not included.

3 - The three first levels only |
The three first levels of the complete item designation, counted from left to right, are included in the numbering area definition. Any levels lower than that, if they exist, are not included.

4 - The four first levels only |
The four first levels of the complete item designation, counted from left to right, are included in the numbering area definition. Any levels lower than that, if they exist, are not included.
